Standard operation, daily maintenance and skills of pipettes

1. Set the pipetting volume

When adjusting from a large volume to a small volume, it is the normal adjustment method, just rotate the scale counterclockwise;

When adjusting from a small volume to a large volume, you can first adjust the scale beyond the set volume clockwise, and then return to the set volume, so as to ensure the best accuracy.

2. Assembly of pipette tips

For a single-channel pipette, insert the pipette tip vertically into the pipette tip, turn it slightly left and right, and tighten it;

It is not advisable to use a pipette to repeatedly hit the tip to tighten. This operation will cause the pipette parts to loosen due to strong impact. In severe cases, the knob to adjust the scale will be stuck.

For multi-channel pipettes, align the first pipette with the first pipette tip, insert it at an angle, and shake it slightly before and after to tighten the pipette tip. After the pipette tip is inserted, it will slightly exceed the O-ring.

3. Maintenance:

1. If the liquid accidentally enters the piston chamber, the pollutants should be removed in time;

2. After using the pipette, adjust the pipette range to the maximum value, and place the pipette vertically on the pipette rack;

3. According to the frequency of use, all pipettes should be washed regularly with soapy water or disinfected with 60% isopropyl alcohol, and then washed with double distilled water and dried;

4. Avoid placing at a higher temperature to prevent liquid leakage or inaccuracy caused by deformation;

5. Find professionals to deal with problems in time.

4. Matters needing attention

1. When the pipette nozzle has liquid, do not place the pipette horizontally or upside down to prevent the liquid from flowing into the piston chamber and corroding the pipette piston;

2. Properly use the pipette to aspirate and discharge liquid to achieve high accuracy.

3. The usual method to check whether there is liquid leakage: stop in the liquid for 1-3 seconds after absorbing the liquid and observe whether the liquid level in the tip drops; if the liquid level drops, first check whether there is any problem with the tip. The liquid level still drops after the tip indicates that there is a problem with the piston assembly, and you should find a professional maintenance person to repair it.

4. Pipettes requiring high-temperature sterilization should first be checked whether the pipette used is suitable for high-temperature sterilization before processing.
