Construction and Application of Protein-DNA Interaction Piezoelectric Array Sensor
[Abstract] Objective To establish a new piezoelectric biosensor detection platform for protein 2 DNA interaction and apply it to the real-time detection of NF2κB p65 protein. Methods AT-cut type fundamental frequency 10MHz gold-plated square quartz crystal wafer was used as the sensing element, and a 2 × 5 type sensor detection microarray was fabricated. The thiol-modified NF2κB cis-acting element sequence probe was fixed on the array by molecular self-assembly technology On the gold membrane electrode of the pool, the concentration of p65 binding subunit of NF2κB was detected in real time. Results The protein 2 DNA sensor constructed under the gas phase conditions had a detection frequency change of ± 1 Hz within 30 min, and the liquid phase detection frequency had a change of ± 2 Hz; as the protein concentration increased, the p65 protein combined with the DNA probe The frequency change of the sensor caused by the reaction gradually increases. When the protein concentration is 40 ng / μl, the response value of the frequency change is the largest. Conclusion A new piezoelectric biosensor detection platform for protein 2 DNA interaction was successfully constructed, and real-time detection of NF2κB p65 was achieved.
